Description
Adsorption buffers are carefully selected and tested for each lectin. Elution buffers are optimized for defined applications and recommended for each lectin. In some cases different elution buffers are available for one lectin. The carbohydrate concentration can be different for high and low affinity interactions or even different sugars are available for an improved elution specificity and efficiency.
The more complimentary the elution sugar is to the lectin binding site the higher is the affinity towards it and the concentration for desorption of the glycoproteins can be reduced. |

Application
Lectin affinity separation of α1-glycoprotein on AffiSep-SNA column. The two chromatograms show elution profiles of α1-glycoprotein using different buffer systems. Elution buffer B2 contains sialyllactose which has higher affinity for SNA than lactose (Elution buffer B1). Thus elution can be achieved in a smaller volume and with a lower sugar concentration using sialyllactose.
